MCLAB's yeast two-hybrid screening service uses commercial available gal4-based YTH systems for identifying protein-protein interactions. We can perform the entire screening, from bait construction through validation of positive clones. - Bait cloning & optimizing screening condition - cloning of the 'bait' sequence into the binding domain plasmid, characterize the bait construct, and set up screening condition
- cDNA library construction and large scale screening - greater than or equal to 1x107 independent clones are screened for their ability to interact with the bait, and up to 100 potential interactors are picked on the basis of their ability to grow on basic medium lacking histidine
- Phenotypic Assessment - additional phenotypic assays, e.g. ß-gal lift assay are used to verify the candidates
- Positive clone validation (e.g.in vitro binding assay, colony sequencing)
- DNA sequencing for screening interesting positive clones
- Full-length cDNA cloning for positive clones
- Domain mapping ( interaction domain)
- Generating antibody against preys
